Le docteur Jon Banks (Jude Law) est un jeune psychiatre ambitieux en quête d'une opportunité pour se mettre en selle. Quand Emily (Rooney Mara), une nouvelle patiente, réclame ses services après une apparente tentative de suicide, il lui prescrit toute la liste des bons vieux régulateurs d'humeur. Mais lorsqu'il teste sur elle un nouveau médicament expérimental appelé Ablixa, elle retourne lentement à la personne qu'elle fut auparavant, heureuse, pleine d'énergie et amoureuse de son mari Martin (Channing Tatum). Soudain, une nuit, Martin est brutalement battu à mort dans leur cuisine.
