Walter is de grootste Muppets fan ter wereld. Samen met zijn beste vrienden Gary (Jason Segel) en Mary (Amy Adams) gaat hij op vakantie in Los Angeles. Tijdens zijn bezoek aan The Muppets Studios ontdekt hij het slechte plan van de rijke oliemagnaat Tex Richman (Chris Cooper). Hij wil The Muppets Studios veranderen in een groot olieplatform. Walter, Mary en Gary gaan daarom op zoek naar The Muppets om ze weer bij elkaar te brengen voor de ultieme Muppet Show "The Greatest Muppet Telethon ever" om zo 10 miljoen dollar op te halen en de Studios te kunnen redden. Maar The Muppets bij elkaar brengen blijkt nogal een lastige opdracht. Ze zijn al jaren niet meer samengekomen en ieder van hen leidt nu een eigen leven. Fozzy zit nu bij een band genaamd Moopets, Miss Piggy is een moderedactrice bij Vogue en woont in Parijs, Animal zit in een kliniek in Santa Barbara om zijn woede-uitbarstingen onder controle te krijgen en Gonzo is een zeer bekende loodgieter geworden.
